RIBOSOMAL RNA SCAFFOLDS FOR PROTEIN-FREE AND TEMPLATE-FREE SYNTHESIS OF PEPTIDES

We have identified two fragments in the ribosome peptidyl center that can synthesize 9-merpoly-lysine. This is the first time that protein-free rRNA ensemble is shown to synthesize oligoswith only Mg2+ ion and water. The peptide synthesizing is template free and proofreading free.Therefore, any amino acids (AA) such as D- AA or beta-AA can be incorporated into an oligochain for drug discovery. Non-natural AAs are novel drug candidates because they areundegradable by cellular proteases. However, due to the high fidelity of ribosome synthesis, it isextremely difficult to incorporate non-natural AA into a peptide chain. But our finding is shortrRNA scaffolds without fidelity checking, making the synthesis of any oligos possible. The rRNAscan be engineered to improve the function.
